Human Cancer Biology GM-CSF Production by Tumor Cells Is Associated with Improved Survival in Colorectal Cancer

چکیده
Purpose: Colorectal cancer infiltration by CD16þmyeloid cells correlates with improved prognosis. We addressed mechanistic clues and gene and protein expression of cytokines potentially associated with macrophage polarization. Experimental Design: GM-CSF or M-CSF–stimulated peripheral blood CD14þ cells from healthy donors were cocultured with colorectal cancer cells. Tumor cell proliferation was assessed by H-thymidine incorporation. Expression of cytokine genes in colorectal cancer and autologous healthymucosa was tested by quantitative, real-time PCR. A tumor microarray (TMA) including >1,200 colorectal cancer specimens was stainedwithGM-CSFandM-CSF–specific antibodies. Clinicopathological features and overall survival

Background: Survival of Colorectal cancer (CRC) patients is considerably stage-dependent therefore, early diagnosis is a pivotal factor in decreasing mortality and morbidity associated with this cancer. GM-CSF and IL-7 are reported to increase in different cancers and we aim to investigate the pre-treatment serum levels of GM-CSF and IL-7 in Iranian patients with colorectal cancer. Methods: ...

Background: Colorectal cancer is the third most common type of aggressive cancers. Chemotherapy, surgery, and radiotherapy are the common therapeutic options for treating this cancer. Due to the adverse side-eff ects of these methods, immunotherapy is considered as an appropriate alternative therapeutic option. Treatment through the application of monoclonal antibodies is considered as a n...
متن کاملTumor and Stem Cell Biology Immune-Dependent and Independent Antitumor Activity of GM-CSF Aberrantly Expressed by Mouse and Human Colorectal Tumors
Granulocyte-macrophage colony-stimulating factor (GM-CSF/CSF2) is a cytokine produced in the hematologic compartment that may enhance antitumor immune responses, mainly by activation of dendritic cells. Here, we show that more than one-third of human colorectal tumors exhibit aberrant DNA demethylation of the GM-CSF promoter and overexpress the cytokine. Mouse engraftment experiments with autol...
متن کاملSerum granulocyte macrophage-colony stimulating factor: a tumor marker in colorectal carcinoma?
BACKGROUND/AIMS Granulocyte macrophage colony-stimulating factor (GM-CSF) is a hematopoietic growth factor, 23 kDA molecular weight with a glycoprotein nature, which is also an immune modulator. The levels of GM-CSF and its role in the pathophysiology of several cancers such as ovarian, breast have been investigated. The aim of the present study was to determine the effect of GM-CSF and carcino...
